Principles of E. coli Protein Expression
Expression vector
Small circular DNA can self-replicate. A complete plasmid vector must have a replication source, a promoter, an inserted target gene, a selection marker and a terminator. Depending on the type of expressed protein, they can be divided into simple expression vectors and fusion expression vectors.
Expression of host bacteria
The selection of host bacteria is mainly based on the characteristics of the host bacteria and the characteristics of the target protein. For example, the target protein needs to form disulfide bonds. Customers can choose the Origami 2 series, which can greatly increase the possibility of disulfide bond formation in the cytoplasm and improve the solubility and activity of the protein.
