Our company's Humanization Approach

Antibodies can be "humanized" by grafting key non-human amino acids onto their antibody framework. This mostly involves the grafting of amino acids into complementarity determining regions (CDRs), but it also involves other amino acids from the framework that are relevant for the VH:VL interface and CDR orientation. Our humanization service strikes a balance between adding as much human components as feasible to lower the danger of immunogenicity and keeping just the right amount of non-human components to preserve the parent antibody's original binding activity.

By integrating humanization with our multi-stage affinity maturation service (STEM TM), our strategy enables us to simultaneously generate superior antibodies. Below is our monoclonal antibody humanization workflow.

(1) Selection of the human germline framework - We integrate the heavy and light chain CDRs of chosen clones into the human framework using bioinformatics analysis. To avoid compromising immunogenicity, we think of amino acids as being tolerated in the human body.

(2) Gene synthesis and recombinant IgG production - We cloned the lead candidate directly into an IgG expression vector and expressed it in HEK293 cells.

(3) Further humanization and affinity maturation using STEM TM - In order to get stronger affinity and specificity, or cross-reactivity to the same antigen in various species for animal investigations, we insert mutations to further humanize the CDR and pan against the target antigen in a progressively strict manner.
