Rodent Dominant Lethal Test

Dominant lethal mutations are mutations that occur in spermatogenic cells and do not cause functional abnormalities in the ligand but can cause the death of the fertilized egg or developing embryo. Our company performs rodent dominant lethal tests on a variety of samples according to the appropriate test standards, and can also provide non-standard testing services for clients based on their needs.

This service provides germ cell mutagenicity testing to detect genetic damage to whole mammalian germ cells and to further confirm positive results obtained from in vitro tests or other testing systems to evaluate whether a sample can cause a dominant lethal mutation.

Test Range

Veterinary drugs, pesticides, chemicals, fungicides, organic toxicants, solid waste, domestic waste leachate, hazardous waste, sewage, sludge, water quality, surfactants, etc.

About Rodent Dominant Lethal Test Service

Test Method

Our company exposes male animals to the test sample by appropriate means and then mates them with uninfected, uncrossed females. After mating, the females are executed in the second half of gestation, the abdominal cavity is dissected, the uterus is removed, and the number of embryos implanted, early stillbirths, late stillbirths, and live fetuses in both uteruses are examined.

The males are mated with another batch of uninfected and unmated females at regular intervals, and so on for several batches to ensure coverage of one sperm cycle.

Sample Preparation

We prefer distilled water as the solvent. If the samples are insoluble in water, we prepare emulsions or suspensions with edible oil, edible starch, and 0.5% sodium carboxymethylcellulose. All samples are freshly prepared before the test.

Test Animals

Animal strains with a low background of dominant lethality, high pregnancy rate, high vegetative population, healthy, and sexually mature should be used.

Our company recommends the use of sexually mature rats or mice.

Control Groups

We set up the appropriate positive control and positive (solvent) control for each sample in each test, and the control animals should be operated on in the same manner as the animals in the stained group, except that the sample is not given a stain.
